MCF 10A 人正常乳腺上皮細胞

描述

MCF 10A細胞株是一個非致瘤的上皮細胞株。 這株細胞通過在低鈣離子濃度的無血清培養(yǎng)液中長時間培養(yǎng)而得到。 MCF 10A是從群體中的貼壁細胞繁衍得到的。也有從懸浮群體繁衍得到的細胞（見MCF 10F, ATCC CRL-10318）。 細胞呈唾液酸粘蛋白陽性, 細胞角蛋白陽性, 牛奶脂肪球抗原陽性。在膠原中表現(xiàn)出三維生長, 并在長滿后形成圓頂狀。 因而, 細胞沒有終端分化或蓑老的表型。這株細胞對胰島素、糖皮質激素、霍亂毒素和表皮生長因子（EGF）有響應。 在電子顯微鏡下特征象燈管細胞而不象肌上皮細胞。 通過MFA-Breast和MC-5單克隆抗體檢測陽性表明它表達乳房特異性抗原。 培養(yǎng)液中的鈣離子濃度對細胞形態(tài)有重要影響。

使用Clonetics公司生產(chǎn)的MEBM基本培養(yǎng)液和SingleQuot添加劑（ATCC不使用正大霉素-兩性霉素B）配成的MEGM（目錄號CC-3150）, 添加100鈉克/毫升霍亂毒素。請不要過濾EGF。
