SIRPα(Signal Regulatory Protein α)是一種免疫球蛋白超家族跨膜蛋白,主要表達(dá)于 髓系細(xì)胞(如巨噬細(xì)胞、樹突狀細(xì)胞)表面,其胞外區(qū)含三個(gè) Ig 樣結(jié)構(gòu)域(D1、D2、 D3),其中 D1 結(jié)構(gòu)域特異性結(jié)合 CD47;胞內(nèi)段含免疫受體抑制基序(ITIM), 可招募 SHP-1/SHP-2 磷酸酶傳遞抑制信號。SIRPα通過調(diào)控吞噬活性維持自身免疫耐受, 防止巨噬細(xì)胞誤傷正常細(xì)胞。 CD47 是一種五次跨膜蛋白,廣泛表達(dá)于紅細(xì)胞、血小板及腫瘤細(xì)胞表面,其胞外 N 端 IgV 樣結(jié)構(gòu)域與 SIRPα結(jié)合,被稱為“別吃我"(Don’t eat me)信號分子,在紅細(xì)胞穩(wěn) 態(tài)(防止脾臟巨噬細(xì)胞清除年輕紅細(xì)胞)及腫瘤免疫逃逸中起關(guān)鍵作用。 當(dāng) CD47 結(jié)合 SIRPα后,SIRPα胞內(nèi) ITIM 基序被磷酸化,招募 SHP-1/SHP-2 磷酸酶, 通過去磷酸化作用抑制下游促吞噬信號。 | |
